Candidacidal Activity of Mouse Peritoneal Macrophages Activated with BCG Immunoadjuvants in Vivo and in Vitro



Abstract
Peritoneal macrophages obtained from BCG adjuvant-immunized mouse and normal mouse were activated with either BCG, Corynebacterium parz,um or Candida albicans adjuvant in vitro and then were infected with C. albicans. The phagocytic and nonphagocytic candidacidal activities were quantitated by microscopic examinations of Giemsa-stained macrophages and methylene blue-stained extracellular candida cells.
The macrophages obtained either from BCG adjuvant-immunized animals or by in vitro activation of normal macrophages with BCG adjuvants exhibited enhanced phagocytic and candidacidal activities. Extracellular candidacidal acitvities of macrophages obtained from BCG adjuvant-immunized animals were abolished by reactivation in vitro with bacterial adjuvants.
The incorporation of mouse anti-candida ascitic fluids into the culture medium also enhanced the phagocytic and intracellular candidacidal activities of normal macrophages, whereas the treatment of normal macrophages with BCG adjuvants in vitro enhanced the extracellular candidacidal activities .
These results suggest that macrophages could be activated by either spcific or nonspecific immunization and production of opsonic antibodies by immunizations with specific antigens may be required for the control of candida infections.
